Orthogonal validation of CRISPRCas9 and siRNA generated phenotypes using cell painting

This application note explores the cell cycle and its key regulatory proteins AURKB, GMNN, and PLK1 using CRISPR-Cas9 and siRNA techniques. High-content imaging and analysis are used to compare phenotypes resulting from gene knockouts/knockdowns. Both methods led to significant cellular changes, highlighting these proteins' importance in cell division and potential as therapeutic targets. Principal Component Analysis revealed differences between siRNA and CRISPR-Cas9 effects, particularly for GMNN inhibition.
